Contrary to popular belief, and despite obvious similarities, the Dionne Quintuplets do not play "themselves" in this film, but the fictional "Wyatt Quintuplets;" Jean Hersholt does not play Dr. Dafoe, but the fictional Dr. John Luke.

Filming of The Dionne Quintuplets took place at their home in Callander, Ontario, for only one hour per day between 10:30 and 11:30 AM, under the technical supervision of Dr. Allan R. Dafoe. The crew could set up the props and cameras within 10 minutes, and the girls were quickly dressed in sewn-together panties, petticoats and dresses, fastened with a single zipper. They were taught a bit of English to sing the song "All Mixed Up," written for the film. Their salary for the 16-day shoot was $100,000. The scene with the puppies was unrehearsed so their surprise and delight could be photographed.

The three nurses attending the quintuplets at the birthday party, Sigrid Ulrichson, Louise Corriveau and Mollie O'Shaughnessey were some the quintuplets' actual nurses.
